No. They don't. In spite of all the things you pointed out, in spite of the fact that she all but is twirling a handlebar mustache, in spite of the fact that she's so obvious that a genre savvy person would dismiss her as antagonist on the grounds that she's TOO obvious, no one thinks she's the Big Bad. In fact, the TV Tropes page for this series has this:

Wham Episode - For starters, Neferet is portrayed as quite likeable with nothing suspicious hinted about her in Marked. In Betrayed, she's revealed to be the Big Bad. For the rest of the series to come. Wham.

Yeeeeah.

Of course, the vampires in general seem pretty insensitive to the suffering and odd behavior of others. I really am not sure if it's supposed to be a deliberate character flaw, or if it's just bad writing. The Fledgeling Handbook 101, for example, tells about how Boudicca's daughters ended up becoming vampires and attending a Ye Olde House of Night after their people were defeated, they were repeatedly raped, and their mother committed suicide in front of them. Apparently none of the other vampires thought that they'd, y'know, be hurting from this. The story ends with the girls founding a student group to provide emotional support to others which suggests that yes, we are supposed to see vampires as sometimes being unfeeling.

Except that still doesn't explain why people who had lived in human society until several weeks ago fail to realize that someone is obviously off their rocker.

And yes, the Cast ladies went there. Unbelievable, no?
